摘要 PROBLEM TO BE SOLVED: To measure the melting point of a two-chain nucleic acid in the state where the nucleic acid is bonded to a substrate, or the melting point of each two-chain nucleic acid corresponding to each probe of a nucleic acid chip immobilized on a plurality of probes. SOLUTION: The first nucleic acid of a probe is immobilized onto the substrate surface, and formation or dissociation depending on the temperature of a hybrid acquired from the first nucleic acid on the substrate surface and the second nucleic acid hybridizable therewith is measured by utilizing fluorescence labeled on the second nucleic acid by an optical system focusing onto the substrate surface, to thereby measure the melting point of the hybrid comprising the nucleic acids.
