A composition for inhibiting activity of beta-secretase(beta-site amyloid precursor protein cleaving enzyme, BACE) by using grape seeds is provided to prevent dementia without side effects by inhibiting activity of beta-secretase. The composition for preventing dementia comprises the aqueous extract or alcohol extract of grape seeds as an effective ingredient, wherein the aqueous extract or alcohol extract of grape seeds specifically inhibits activity of beta-secretase relating to beta-amyloid peptide production; the aqueous extract or alcohol extract of grape seeds contains proanthocyanidin as an oligomer and polymer of polyhydroxy flavan-3-ol units; and the composition is formulated as fermentation oil, beverage or health food.
