PROBLEM TO BE SOLVED: To provide a microscope using an oil immersion system objective lens capable of eliminating spontaneous fluorescent light from an immersion oil filling between a sample and the oil immersion system objective lens as much as possible, thereby enhancing the SN of a fluorescent image, and to provide an observation method of the microscope using the oil immersion system objective lens. SOLUTION: The microscope includes an oil immersion system objective lens 1 and an imaging lens 2, for observing a sample by filling a space with oil between a cover glass CG protecting the sample and a lens face nearest to the sample. The front focal distance and the front focus position are controlled in such a manner that a beam is focused at a predetermined depth position of the sample while a work distance WD between the cover glass CG and the lens face nearest to the sample in the oil immersion system objective lens 1 satisfies 0<WD≤0.03 mm, by moving a lens group G2, G3, G4 disposed in an image side than the lens nearest to the sample in the oil immersion system objective lens 1 along the optical axis.
