A bio-sensor is provided to be able to minimize the blood amount required for measurement and also be easily manufactured. The bio-sensor comprises: an insulating lower substrate(30) which includes at least one electrode(31) and has an enzymatic reaction layer(33) formed on the electrode to react with an analyte; an upper substrate(10) which faces with the lower substrate, is composed of a conductive material and has a line resistance of less than 3kФ/mm; and an adhesive plate(20) which forms a sample introducing portion(21) on the enzymatic reaction layer with certain height and combines the lower substrate with the upper substrate. In the bio-sensor, one side end of the upper substrate acts as an electrode(41) where an electron transport medium material contained in the enzymatic reaction layer is oxidized or reduced, and the other side end of the upper substrate acts as an electricity contact portion(40) which electrically contacts with a measuring machine. The enzymatic reaction layer includes at least one kind of enzyme and at least one kind of electron transport medium material.
