PROBLEM TO BE SOLVED: To provide a method for purifying PrPres from a biological sample to be used for qualitative and/or quantitative determination of the PrPres contained in the sample. SOLUTION: The method essentially consists in: (1) a step for incubating the biological sample with a protease or a buffer solution A comprising at least one kind of a surfactant in a prescribed amount, to form a suspension S1; (2) a step for adding to the obtained suspension S1 a buffer solution B in a prescribed amount sufficient for forming phase separation without solubilizing the PrPres, and consisting of a prescribed alcohol or mixture thereof; (3) a step for centrifuging the suspension resulting from the step (2); (4) a step for collecting thin film present at the interface; (5) a step for resolubilizing the thin film by the buffer solution A; (6) a step for centrifuging the obtained residue; and (7) a step for solubilizing the obtained residue in a buffer solution C containing the surfactant in the step (1), a chaotropic agent or a mixture thereof.
