<p>It is intended to enable the synthesis of a saccharide derivative (for example, a polysaccharide, a glycolipid, a sugar amino acid, etc.) designed depending on the purpose at a low cost and a high yield by the phase dissolution/phase separation liquid phase synthesis method. By using a temperature-dependent phase dissolution/phase separation solvent prepared by mixing methylcyclohexane (MCH), which is a solvent having a low polarity, with propionitrile (PN), which is a solvent having a high polarity, as a reaction solvent, a sugar donor (D) having a sugar skeleton (S) and carrying a leaving group (E) introduced into a binding site and a receptor (R) having a chemical modification (Q) and carrying a hydroxyl group (F) at the binding site are selectively dissolved in the respective solvents. While maintaining the reaction solvent in the phase dissolution states, the sugar skeleton (S) of the sugar donor (D) is bonded to the chemical modification (Q) of the receptor (R) in the liquid phase to thereby synthesize a saccharide derivative (Y) selectively soluble in methylcyclohexane. Subsequently, the reaction solvent is converted into the phase separation state to thereby separate methylcyclohexane (MCH) in which the saccharide derivative has been dissolved.</p> |
