PROBLEM TO BE SOLVED: To provide a method for producing a protoplast of a yeast belonging to the genus Sporobolomyces. SOLUTION: The protoplast is produced by culturing the yeast belonging to the genus Sporobolomyces under a culture condition of inhibiting the synthesis of a cell wall, and adding the cultured product to an enzymically treating solution of the cell wall, containing two or more kinds of cell-wall digesting enzymes.
