NOVEL NON-HUMAN ANIMAL MODEL FOR ALZHEIMER'S DISEASE HAVING DETERIORATED A-ß EXCRETION FUNCTION |
摘要 |

Disclosed is a novel non-human animal in which ß-amyloid peptide (A-ß) is produced but the metabolism/excretion of A-ß is impaired and which can be used particularly as a model of sporadic Alzheimer's disease. A transformed non-human animal having deteriorated A-ß metabolism/excretion, which is produced by causing the deletion of a gene encoding a-tocopherol transfer protein (aTTP) in a non-human animal and administering A-ß to the animal; or a transformed non-human animal having deteriorated A-ß metabolism/excretion, which is produced by causing the deletion of a gene encoding aTTP in a non-human animal having mutations in at least one gene selected from the genes encoding amyloid peptide precursor protein (APP), presenilin 1 (PS1) and presenilin 2 (PS2), respectively. Since, in this animal, endogenous A-ß is produced and accumulated in the brain to develop AD actually, the animal is the first-ever model animal for the study of the AD development mechanism.
|
