<p>Tetrahydroquinoline-based thiazolidinediones(TZD) derivatives having specific function as PPARgamma ligand and a preparation method thereof are provided to reduce blood glucose level by promoting the transcription activation of PPARgamma with reduced liver toxicity and treat diseases associated with activity of PPARgamma including cancer and dementia. The tetrahydroquinoline-based thiazolidinediones derivatives represented by the formula(1) or pharmaceutically acceptable salts thereof are provided, wherein R is C1-C20 alkyl, substituted alkyl or aromatic ring. The preparation method of the tetrahydroquinoline-based thiazolidinediones derivatives comprises the steps of: (a) hydrogenating(compound 1) and esterifying(compound 2) quinaldinic acid, introducing alkyl group into nitrogen of tetrahydroquinoline(compound 3), and reducing the compound 3 to prepare compound 4; (b) condensing 4-hydroxybenzaldehyde with 2,4-thiazolidinedione to prepare compound 9, hydrogenating the compound 9 to prepare compound 10, acetylating the compound 10 to prepare compound 11, introducing a protecting group into the amine group of the compound 11 to prepare compound 12, and deacetylating the compound 12 to prepare compound 13; and (c) reacting the compound 13 with the compound 4 to prepare compound 14, and removing the amine-protecting group from the compound 14 to prepare compound 15.</p> |
