Disclosed are a reagent and a kit usable in a coagulation system assay of a direct factor Xa inhibitor and a method for the assay (the term "coagulation system assay" means a method for quantification of the effect of a coagulation factor inhibitor on the coagulation system or a method for determination of the concentration of a coagulation factor inhibitor in the blood or plasma). A reagent or the like for use in a coagulation system assay which comprises factor Xa, a phospholipid and a calcium ion; that is, a reagent or kit useful in a coagulation system assay of a direct factor Xa inhibitor which comprises factor Xa, a phospholipid and a calcium ion, provided that any reagent or kit additionally including factor V or Va or a factor V activating enzyme is an exception to the reagent or kit, and a method for coagulation system assay of a direct factor Xa inhibitor using the reagent or kit.
