A hydrogen sensor is based on the detection and measurement of a reaction product by the consumption of hydrogen via a redox reaction using a hydro genase and an electron acceptor in solution. The detection is carried out using a sensing element disposed proximate to or in the solution. The amount of hydrogen present is measured via a signal that results from a change in the concentration of the reduced form of the electron acceptor, which has been correlated to the amount of hydrogen in contact with the solution. The measurement can be that of an oxidation current resulting from the redox reaction or of the optical intensity of a product or reactant in the redox reaction. The detector can be coupled to a processor with memory to store calibration data between the oxidation current or optical intensity measured and the H2 concentration and process the signal from the detector to output a hydrogen concentration. The sensing element and the reagent well can be in an integrated chip-based microfluidics sensor.
