FIELD: molecular biology, biochemistry. ^ SUBSTANCE: invention proposes a method for amplification of nucleic acid (NA) involving carrying out a primer reaction elongation on matrix in the presence of at least one nucleotide as a precursor of DNA recognized by a cleaving agent, digestion of elongated primer but retaining a bind with matrix NA by indicated agent to form fragments comprising free OH-group at 3'-end and elongation of prepared fragment on matrix. The distinctive feature of a method is the combining all reactions in a single vessel and using the reaction mixture containing all necessary components, and DNA polymerase, a cleaving agent and the reaction conditions are selected by taking into account their simultaneous functioning.
