摘要 The invention relates to a method for preparing a synthetic fragment of nucleic acids, in which a large first fragment of double strand synthetic DNA of a determined sequence comprising a catemer concatenation in a determined order of a plurality of n second small adjacent synthetic DNA fragments is prepared. Said method comprises a first PCR-type amplification reaction step for a series of n oligonucleotides of determined sequences, without exogenous primers, in temperature conditions which enable the hybridisation of said oligonucleotides, followed by an elongation of the complex obtained, to be placed end-to-end in a determined order of said oligonucleotides, the sequences of said oligonucleotides corresponding successively and alternatively to the sense and antisense sequences of the different said second synthetic fragments, and each oligonucleotide containing, in the regions 5' and 3' thereof, sequences which are complementary to those of the following and preceding oligonucleotides, if necessary. The inventive method also comprises a second step for amplification by means of specific primers of the ends 5' and 3' of said direct strand of said large first synthetic fragment to be prepared, enabling identical copies of the large first fragment to be produced.
