<p>A gene detection method whereby a mutation at a specific base in a target gene or the presence or absence of the same is determined. This method comprises: (i) amplifying each of a target gene sample and a control gene sample, which carries a wild type or standard type base sequence for the target gene, by a PCR with the use of a pair or primers involving a primer having an RNA polymerase promoter sequence at the 5'-end; (ii) subjecting the double-stranded DNA obtained by the above PCR to an in vitro transcription reaction to thereby form a single-stranded RNA; (iii) hybridizing the single-stranded RNA as described above with a fluorescent-labeled probe, wherein a fluorescent dye is bonded to a single-stranded DNA comprising a sequence being complementary with at least a part of the base sequence of the control gene as described above, to thereby form an RNA/DNA hybrid; and (iv) comparing the fluorescent intensity of the RNA/DNA hybrid derived from the target gene sample with the fluorescent intensity of the RNA/DNA hybrid derived from the control gene sample.</p>
