PROBLEM TO BE SOLVED: To provide a method for separating and purifying a nucleic acid by adsorbing a nucleic acid in a biomaterial on a solid-phase surface, cleaning and then eliminating a nucleic acid, with which more biomaterials are treated without prolonging a treatment time for obtaining a solution to adsorb nucleic acid on a solid phase. SOLUTION: In the method for separating and purifying a nucleic acid comprising (1) a process for adding a solution to a biomaterial to prepare a sample solution containing a nucleic acid, adding a water-soluble organic solvent or a solution containing a water-soluble organic solvent to the sample solution to prepare the sample solution containing the water-soluble organic solvent, (2) a process for bringing the sample solution containing the water-soluble organic solvent into contact with a solid phase to adsorb the nucleic acid on the solid phase, (3) a process for bringing a cleaning liquid into contact with the solid phase and cleaning the solid phase in a state of a nucleic acid being adsorbed on the solid phase and a process for bringing a recovery liquid into contact with the solid phase and eliminating the nucleic acid from the solid phase, in the process (1), the water-soluble organic solvent or the solution containing the water-soluble organic solvent is added to the sample solution dividedly at least twice.
