2'-Deoxyguanosine compounds represented by formula (2), may be prepared by desulfurizing an 8,2'-anhydro-8-mercapto-9-beta-arabinofuranosylguanine compound represented by formula (1) in a solvent in the presence of a base using nickel or a nickel alloy. wherein R¹, R², R⁴, and R⁵ are each independently a hydrogen atom or a hydroxyl-protecting group, and R³ and R⁶ are hydrogen atoms or amino-protecting groups.
