<p><P>PROBLEM TO BE SOLVED: To provide a method of nucleotide sequencing of a polynucleotide. <P>SOLUTION: Disclosed is a method of nucleotide sequencing of a polynucleotide, wherein the method comprises the following steps: (a) supplying a polynucleotide and a probe having a nuclease recognition site; (b) blocking the nuclease recognition site on the polynucleotide; (c) ligating the probe to the end of the polynucleotide; (d) identifying one or more nucleotides at the end of the polynucleotide; (e) cleaving the polynucleotide with a nuclease recognizing the nuclease recognition site of the probe such that the polynucleotide is shortened by one or more nucleotides; and (f) repeating steps (c) through (e) until the nucleotide sequence of the polynucleotide is determined. <P>COPYRIGHT: (C)2006,JPO&NCIPI</p> |
