发明名称 METHOD FOR PRODUCING DNA POLYMERASE
摘要 PROBLEM TO BE SOLVED: To provide highly heat-resistant DNA polymerase having 3'-5' exonuclease activity being a calibration function of DNA chain elongation and a strong primer elongation activity. SOLUTION: Disclosed is a thermophilic archaebacteria-derived heat-resistant heterodimer enzyme featured by that the enzyme consists of a small subunit and a large subunit having molecular weight of about 70±7 kDa and about 144±14.4 kDa respectively, the large subunit does not contain an intein sequence, at least about 50% of activity is maintained after heating treatment at 85°C for 1 hr, at least about 20% of activity is maintained after heating treatment at 90°C for 1 hr, and the enzyme has DNA polymerase activity and 3'-5' exonuclease activity.
