PROBLEM TO BE SOLVED: To provide a method for forming sub-pools used for testing the presence of a specific agent in a fluid sample. SOLUTION: This method comprises (a) associating fluid samples to the matrix elements of a N-dimensional matrix, respectively, identifying the matrix elements with N index values, respectively, corresponding the N indexes to the N dimensions of the matrix in a ratio of 1:1; (b) forming N aliquots from above-mentioned samples; and (c) associating the aliquots to form sub-pools, wherein each of the sub-pools contains one aliquot from the above-mentioned fluid samples related to the matrix elements having identification values on one of the N indexes.
