<P>PROBLEM TO BE SOLVED: To isolate a D-aminoacylase gene derived from Streptomyces thermonitrificans CS5-9 strain; and to provide a method for producing D-aminoacylase by using the gene. <P>SOLUTION: A primer synthesized by comparing the well-known D-aminoacylase gene is constructed, and amplification fragments are obtained by a PCR method. The target D-aminoacylase gene derived from the Streptomyces thermonitrificans CS5-9 strain is isolated by carrying out genome southern hybridization by using a part of the amplification fragments as a probe, and cloning the hybridized plasmid having 6.5 kbp SacI fragment by a colony hybridization.
