The instant invention relates to a method for amplifying a genomic DNA sequence, comprising at least steps of: i. contacting said genomic DNA sequence with a set of pairs of degenerate oligonucleotide primers hybridizing with primers binding sites of said genomic DNA sequence, each of these sites being homologously present in at least two orthologous forms of said genomic DNA sequence, ii. subjecting the mixture obtained at step i) to a touchdown polymerase chain reaction (PCR) under suitable conditions for obtaining said amplified DNA sequence.
