PROBLEM TO BE SOLVED: To solve problems of a conventional inspection in which conventional gene diagnosis of hematopoietic organ tumor was custom-made and realize ready-made gene diagnosis to general clinical test. SOLUTION: IgH gene forms a V-D-J conjugate (CDRIII) after reconstruction in a basic VDJ region. In order to detect the V-D-J conjugate, JH probe (LJH, VLJH) against J region is used in Southern method and Semi-nested PCR method using a primer FR3A against V region and primers LJH and VLJH against J region is used in PCR method. TCR gene forms a V-D-J conjugate (V-delta 1-D-J delta 1) after reconstruction in a basic VDJ region. In order to detect the V-D-J, a Nested PCR method, in which a primer D3 against V delta 1 in the V region and a primer D4 against J delta 1 in J region and a primer D1 against the outside of the V region and a primer D6 against outside of J region are used, is used. As a result, automation of an inspection method and inspection kit can be completed and ready-made gene diagnosis is realized in the field of general clinical test. COPYRIGHT: (C)2006,JPO&NCIPI
