A method for preparing a biosensor, which comprises providing a substrate having an electrode, applying a solvent containing a mediator, a surfactant, a buffer and a layered inorganic compound on the surface of the electrode, to thereby form an inorganic gel layer for preventing the natural oxidation of the above mediator, and further, forming an enzyme reagent layer containing an oxidoreductase on the above layer; and a biosensor prepared by the method. In the biosensor, due to the above inorganic gel layer, the mediator having been reduced by the reaction of a material to be measured and the oxidoreductase can be measured electrochemically, without being re- oxidized by dissolved oxygen or the like. The above biosensor allows the avoidance of the influence of oxygen to a mediator and the measurement of a substance to be measured in a sample solution with rapidity, with ease and simplicity, with high accuracy.
