METHOD OF AMPLIFYING AND DETECTING NUCLEIC ACID

It is intended to provide a nucleic acid amplification method whereby a mutant gene contained in a minor amount in a sample, which contains a large amount of a wild type gene, can be specifically amplified. Namely, a method of specifically amplifying either a wild type gene or a mutant gene which are both contained in a sample, comprising (a) the step of preparing a primer set containing at least one primer having sequences being homologous or complementary to two or more regions on the gene to be amplified as described above and at least one primer having sequence(s) being homologous or complementary to one or more regions on the gene, wherein a nucleotide residue concerning the mutation is contained at least one of the above-described regions; (b) the step of performing a nucleic acid amplification reaction by using the above-described primer set with the use of a nucleic acid molecule contained in the above-described sample as a template; and (c) the step of performing the subsequent nucleic acid amplification reaction by using the above-described primer set with the use of the amplification product obtained by the former nucleic acid amplification reaction as a template.
