A method of monitoring activity of an enzyme utilises a substrate tagged with a redox-active group, wherein the substrate is incubated with the enzyme to allow modification of the substrate, and the modification is electrochemically monitored by means of the redox-active label. The method may be used to monitor activity of enzymes such as kinases, phosphatases and proteases, as well as oxidoreductases. Preferably the substrate is a peptide labelled with a ferrocenem a fullerene or a quinone. The method may also be used to assay candidate substrates for an enzyme, or candidate inhibitors of the enzyme. |
