METHOD FOR PREPARING MUTANT PROTEIN BY CONTROLLING GENE MUTATION FUNCTION OF CELL

PROBLEM TO BE SOLVED: To establish a cell strain temporarily stopping the expression of an AID (activation induced cytidine deaminase) governing the mutation functions once and then isolating the objective clone in order to isolate the clone producing a useful mutant protein and permanently produce the useful mutant protein.

SOLUTION: The cell comprises an AID gene sandwiched between two loxP sequences in the mutual reverse directions. The change from promotion (ON) to stop (OFF) or OFF to ON in expression of the AID gene present on the upstream side of a region sandwiched between the two loxP sequences and controlled with a promoter can be carried out by reversing the direction of the AID gene when the expression of a Cre (Cre recombinase) is induced by stimulation from the outside of the cell. That is, the ON state of the expression of the AID gene is maintained when the AID gene faces to the forward direction to the promoter. The OFF state of the expression thereof is maintained when the AID gene faces to the reverse direction to the promoter.
