PROBLEM TO BE SOLVED: To provide a method for detecting intracellular micronuclei by solving problems such that, since in a conventional detection method, procedures are complex and there are differences among testing persons, it is difficult to establish standardization covering testing institutions, and also on considering an automatic measurement, there are difficulties in quantitativeness and reproducibility since it is not easy to obtain the distinction between a nucleic region and cytoplasmic region by a conventional staining method. SOLUTION: This method for detecting the intracellular micronuclei is characterized by forming a mother cell obtained by fusing a gene encoding single or a plurality of nucleus-associated proteins with a gene encoding a fluorescent protein, introducing the fused gene into cultured cells for expression, treating the cell for the toxicity or mutagenicity testing of a substance to be tested, performing a limited excitation so that the micronuclear region emits fluorescent light limitedly and measuring the amount of the fluorescent light in accordance with a nucleus-associated protein component quantitatively for enabling the detection of the micronuclei in live cells without performing the fixation of the cells as measuring subjects.
