PROBLEM TO BE SOLVED: To provide a manufacturing method of an optically active 1-substituted-2-methyl-1-butanone which permits the manufacture in a high yield with a high optical purity by a simple and inexpensive method. SOLUTION: (R) or (S)-2-methylbutyric acid is treated with a chlorination agent to give (R) or (S)-2-methylbutyric acid chloride, which is reacted with N-methylmethoxyamine to give N-methoxy-N-methyl-(2R) or (2S)-methylbutanamide, which is subjected to a Grignard reaction with a Grignard agent or a reaction with an organometallic reagent to give a 1-substituted-(2R) or (2S)-methyl-1-butanone.
