摘要 PROBLEM TO BE SOLVED: To provide a method for rapidly and conveniently measuring a total number of Vibrio bacteria. SOLUTION: This method for measuring the total number of the bacteria of a bacterial species of the Vibrio comprises preparing a calibration curve by subjecting the Vibrio bacteria in bacterial concentrations stepwise diluted to real time PCR (polymerase chain reaction) through a TaqMan procedure and, at the same time, subjecting an extract of a specimen comprising a measuring target of the number of the bacteria to the real time PCR through the TaqMan procedure in a similar manner, calculating a bacterial concentration of the bacterial species of the Vibrio in the extract of the specimen, and calculating the total number of the bacteria of the bacterial species of the Vibrio existing in the specimen based on the bacterial concentration. Further, base sequences of two or more of different strains of the bacterial species of the Vibrio are determined with respect to a DNA region which is species-specifically conserved in the bacterial species of the Vibrio comprising the target among ToxR genes but not conserved in other bacterial species of the Vibrio, when the real time PCR is conducted, so that a position of polymorphism existing in the gene is determined and positions of a primer and a probe for the real time PCR are determined so as not to contain the polymorphism.
