TISSUE-SPECIFIC TRANSCRIPTION BY RNA POLYMERASE III

The invention provides materials and methods which enable the tissue-specific transcription of desired DNA sequences by RNA polymerase III. Transcriptional regulatory elements which direct tissue specific transcription by RNA polymerase II are combined with transcriptional regulatory elements from promoters which direct transcription by RNA polymerase III, to achieve tissue specific transcription by RNA polymerase III. Constructs comprising such hybrid promoters are particularly useful for expression of RNA molecules such as siRNAs and ribozymes.
