An apparatus for separation and purification, comprising extraction distillation column (4) for separation and purification of, for example, butadiene; impurity concentration sensors (32,34) for detecting the concentrations of specified impurities other than butadiene; a target matter concentration sensor for detecting the concentration of butadiene in the extraction distillation column; and pressure difference sensor (30) for detecting any pressure difference between column top and column bottom in the extraction distillation column (4). Further, there is provided a relevant method of separation and purification. With the use of the above sensors, the estimated values for pressure difference between column top and column bottom, butadiene concentration at column top and concentrations of specified impurities at the passage of a given period of time are calculated, and control by concentration estimation control means (60) is carried out on the basis of the estimated values. Raw material flow rate control valve (21a) for controlling of the amount of raw material fed to the extraction distillation column (4) and load detection means (61) for detection of the load on the extraction distillation column are provided, and the raw material flow rate control valve (21a) is controlled by load control means (62) in accordance with the detected values obtained by the load detection means (61).
