A method of preparing an extract from cultured mammalian cell, comprising at least the step of rapidly freezing cultured mammalian cells suspended in a liquid for extraction; an extract from cultured mammalian cell prepared by the process; and a method of cell-free protein synthesis using the extract. Preferably, the method of cell-free protein synthesis comprises the step of prior to conducting of synthetic reaction, effecting thermal insulation of a cell-free protein synthesis reaction liquid mixture in a state of containing components other than foreign mRNA for a specified period of time.
