Selective determination of active fluorophores allows, by use of lenses (L6) focussing of the illumination beam on selected planes /optical depths of the object and detection of both the total photoluminescent output and the decay luminescence component, where the focussing lenses may be rotated, tilted or pushed into the light path, is new. An independent claim is also included for an arrangement for illuminating and measuring both total photo-luminescent output and the decay luminescence component and allowing a correction to be made for the latter constant contribution.
