METHOD FOR CULTURING MESENCHYMAL STEM CELL DERIVED FROM HUMAN BONE MARROW BY USING HUMAN SERUM CULTURE MEDIUM

PROBLEM TO BE SOLVED: To provide a method for safely and efficiently culturing a mesenchymal stem cell for transplanting to human from human bone marrow liquid.

SOLUTION: The method for preparing the human mesenchymal stem cell for collecting human bone marrow liquid, culturing the mesenchymal stem cell derived from bone marrow and transplanting the stem cell to human comprises (i) a step for adding 80-120%v/v heparin/buffer solution in the concentration of about 5-15U/mL to quantity of the collected bone marrow liquid, (ii) a step for culturing the bone marrow liquid to which the heparin/buffer solution obtained in the step (i) is added in an α-MEM culture medium containing 10-20%v/v human serum and exchanging the culture medium twice during culture to remove a hemocyte ingredient floating in the culture solution and (iii) a step for selectively releasing a cell group mainly containing the mesenchymal stem cell from a culture vessel by using a cell-dissociating agent, separating an adhesive cell other than the mesenchymal stem cell from the released cell group and selectively culturing the mesenchymal stem cell in other vessel.
