Epothilone analogues represented by Formulas I-IX useful as microtubule stabilizing agents and in the treatment of abnormal cellular proliferation diseases and disorders are disclosed. Methods of making the compounds, compositions containing the compounds, 3D binding models of the binding of epothilone analogues on a,ß-tubulin, and methods for its use in predicting, designing, or selecting therapeutically useful epothilone analogues are also provided.
