PROBLEM TO BE SOLVED: To provide a recombinant cell obtained by a method for site-specifically inducing at least one break of a double strand to a chromosome DNA.

SOLUTION: The invention provides a separated DNA encoding an I-SceI enzyme. The DNA sequence can be introduced into a cloning vector, an expression vector, a transformed cell and a transgenic animal. The vector is useful for the gene mapping and the site-specific introduction of a gene. The recombinant cell transformed by a method to induce at least one site-specific break in a chromosome DNA and inserting a DNA encoding a polypeptide enables (a) the preparation of a cell having a double-stranded DNA having an I-SceI restriction site, (b) the transformation of the cell with at least one plasmid having a DNA encoding I-SceI and (c) the selection of a cell having at least one induced break on double strand.
