摘要 PROBLEM TO BE SOLVED: To provide a method for determining whether or not a patient has a neoplastic lesion and provide a method for determining a neoplastic lesions by using the method. SOLUTION: In order to discriminate neoplastic lesions, a method for determining monoclonality of reconstructed genes is utilized that comprises following five steps (1) to (5): (1) A genome DNA is cleaved with a restriction enzyme (a) that shows high occurrence rate in the genome and recognizes an area site that is not removed off by the reconstruction of a target gene; (2) A linker is connected to the cleaved site by the restriction enzyme of the DNA fragment obtained in (1); (3) A linker-bearing DNA fragment obtained in (2) is cleaved with a restriction enzyme (b) that shows low occurrence rate in the genome and recognizes an area site that is not removed off by the reconstruction of the target gene; (4) A primer that hybridizes on the downstream area of the linker-bearing DNA fragment and another primer that hybridizes on the base sequence that is complementary to the base sequence on the linker area are used to carry out a PCR (polymerase chain reaction) operation by using the fragment obtained in (3) as a template; and (5) The monoclonality of the PCR product that is obtained in (4) is measured whereby the monoclonality of the reconstructed gene is discriminated.
