It is intended to provide a method of producing a dipeptide or a dipeptide derivative characterized by comprising: supplying to an aqueous medium a substance selected from the group consisting of a phosphate donor, adenosine-5'-monophosphate, adenosine-5'-diphosphate and adenosine-5'-triphosphate, a protein having a polyphosphate kinase activity, a culture of cells capable of producing the protein or a processed product of the culture, a protein having an activity of ATP-dependently forming a dipeptide or a dipeptide derivative from one or more amino acids or amino acid derivatives, a culture of cells capable of producing the protein or a processed product of the culture, and one or more amino acids or amino acid derivatives together; forming and accumulating the dipeptide or dipeptide derivative in the aqueous medium; and harvesting the dipeptide or dipeptide derivative from the aqueous medium.
