PROBLEM TO BE SOLVED: To provide an observation method and an observation device for clearly and rapidly detecting the shape characteristics of horny layer cells, above all, the arrangement regularity of the layer cells, and especially the joining condition of the layer cells, even on a non-dyeing condition. SOLUTION: This non-dyeing observation method for horny layer cells is characterized in that: a light transmissive adhesive support body with an adhesive surface is pressure-attached to a skin surface and then exfoliated therefrom; horny layer cells are collected onto the adhesive surface; a light non-transmissive support body is disposed on the adhesive surface side of the adhesive support body; rays of light are projected from the non-adhesive surface side of the support body onto the adhesive surface side thereof by light sources 4 provided around a camera 6 for imaging the layer cells; reflected light thereof is condensed through a polarizing plate 5; the layer cells are imaged by the camera 6; and a picture image acquired is displayed as an enlarged image on a personal computer to observe the state of the layer cells.
