Specific aryl phosphate nucleoside derivatives show activity against HIV without undesirable levels of cytotoxic activity. In particular, these derivatives are potent inhibitors of HIV reverse transcriptase. Examples of aryl phosphate nucleoside derivatives include aryl phosphate derivatives of d4T having one or mote substituents on the aryl group selected from the group consisting of: 3-N(CH<SUB>3</SUB>)<SUB>2</SUB>; 2,6-(CH<SUB>3</SUB>O)<SUB>2</SUB>; 4Br-2-Cl; and 2,5-Cl<SUB>2</SUB>, and wherein the phosphorus of the aryl phosphate group is N-linked to an methyl or ethyl ester of an amino acid residue such as a methoxyalaninyl group.
