PROBLEM TO BE SOLVED: To determine a light illumination position for photo-stimulus in a visual field while performing microscope observation and to accurately photo-stimulate the determined illumination position. SOLUTION: This microscope observation method is equipped with: a step for introducing a substance into a sample A, the substance changing its structure by being illuminated with light of a first wavelength; a step for determining a photo-stimulated part of the sample A by illuminating the sample A with visual light of a second wavelength not changing the structure of the substance while observing the sample A with the substance introduced thereinto through a microscope observation device 14; and a step for illuminating the determined photo-stimulated part with the light of the first wavelength.
