<p><P>PROBLEM TO BE SOLVED: To provide a method for determining a nucleotide sequence of a polynucleotide. <P>SOLUTION: The method for determining the nucleotide sequence of the polynucleotide comprises (a) a step to prepare a probe having a polynucleotide and a nuclease recognition site, (b) a step to block the recognition site of the nuclease on the polynucleotide, (c) a step to ligate the probe to a terminal of the polynucleotide, (d) a step to identify one or more nucleotides on the terminal of the polynucleotide, (e) a step to cleave the polynucleotide by using the nuclease recognizing the nuclease recognition site of the probe to shorten the polynucleotide by one or more nucleotides, and (f) steps to repeat the steps (c) to (e) until the nucleotide sequence of the polynucleotide is determined. <P>COPYRIGHT: (C)2006,JPO&NCIPI</p> |
