PROBLEM TO BE SOLVED: To provide a method enabling, in production of an optical isomer by supercritical fluid chromatography using a mobile phase containing a supercritical fluid and a solvent, reuse of a gas forming the supercritical fluid and stable production of the optical isomer. SOLUTION: The method comprises injecting a sample containing a mixture of optical isomers to a mobile phase containing a supercritical fluid, passing the mobile phase with the sample injected thereto through a column having a filler identifying the optical isomer, and separating and producing a desired optical isomer in the sample from the mixture. In this method, the mobile phase containing the supercritical fluid generated from the liquefied gas and the solvent is generated, the mobile phase passed through the column is separated to the solvent and the gas forming the liquefied gas, the separated gas which has a predetermined concentration or less of the solvent in the separated gas is used as the liquefied gas for the supercritical fluid contained in the mobile phase.
