PROBLEM TO BE SOLVED: To provide a measuring method using an agglutination reaction of an affinitive substance, and a device therefor. SOLUTION: A coupling reaction of the affinitive substance of a measuring object with a coupling partner having coupling affinity with the affinitive substance is measured by the agglutination reaction. The affinitive substance is bonded to a carrier particle, and the coupling partner is agglutinated by the coupling reaction. The method of the present invention includes a process for diluting a reaction liquid containing single particles before counting agglutinated blocks. The dilution process of the present invention makes a bond between the affinitive substance and the coupling partner strong. As a result, the agglutinated block is prevented from being collapsed to enhance measuring sensitivity. The diluted carrier particles and the agglutinated block are discriminated with high precision.
