PROBLEM TO BE SOLVED: To provide a method for analyzing nucleic acids, targeting analysis of such DNA as a higher-order structure DNA. SOLUTION: This method comprises denaturation of a double-strand DNA to its single-strand DNA 1, hybridization of a sequence at a region which is an analytical target to be analyzed and of a genomic DNA, or of primers 3, 4 specific to mutation to the DNA 1, complemental strand synthesis initiated at the primers by using a substrate, polymerase and a single-stranded DNA bonding protein (SSB), resulting chemiluminescence due to pyrophosphoric acid 5 generated in the complemental strand synthesis by using a chemiluminescent agent, and analysis of whether or not mutation in the genomic DNA exists by detecting the luminescence.
