FOLLISTATIN MUTANT POLYPEPTIDE

A follistatin mutant which can specifically inhibit the activity of GDF-8 without inhibiting the activity of activin. The follistatin mutant polypeptide is characterized by (a) containing follistatin domain I, (b) having an amino acid sequence represented by the formula (I): Cys-(X<1>)a-Cys-(X<2>)b-Cys-(X<3>)c-Cys-(X<4>)d-Cys-(X<5>)e-Cys-(X<6>)f-Cys-(X<7>)g-Cys-(X<8>)h-Cys-(X<9>)i-Cys (wherein X<1>, X<2>, X<3>, X<4>, X<5>, X<6>, X<7>, X<8>, and X<9> are the same or different and each independently represents a residue of an amino acid present in the nature, other than cysteine; a is an integer of 2-6; b is an integer of 3-7; c is an integer of 7-11; d is an integer of 0-4; e is an integer of 1-6; f and g each is an integer of 8-12; h is an integer of 4-8; and i is an integer of 11-15) and containing no amino acid sequence having 50% or higher homology to the amino acid sequence of sequence number 2 or 29, and (c) inhibiting the activity of GDF-8 more selectively than the activity of activin.
