PROBLEM TO BE SOLVED: To provide a method for obtaining double stranded DNA by which reaction products having almost uniform size are obtained without the need for many hours and labor in a process where an amplification reaction of double stranded DNA is carried out by using a random primer and the double stranded DNA are fractionated and isolated. SOLUTION: In the proliferation reaction of double stranded DNA 204 by using a random primer 202, at first, the amplification reaction is carried out only one cycle so that the reaction products including a uniform or less sizes may be obtained [treatment (a)]. The reaction product in a reaction liquid is subjected to a fractionation and isolation processes by using a molecular sieve membrane 206 [treatments (b), (c)]. The reaction liquid is refilled [treatment (d)]. These treatment steps (a) through (d) are repeated to obtain a needed amount of the reaction products [treatment (e)] thereby a template DNA of a specific size can be amplified and isolated.
