A method of while suppressing natural coloring, stabilizing a reagent capable of simultaneous quantification of LDL cholesterol and total cholesterol by one-time measuring. There is provided a method of quantifying cholesterol of low-density lipoprotein and total cholesterol contained in a biosample by one-time measuring, comprising the first step of treating lipoproteins other than low-density lipoprotein contained in a biosample to thereby generate hydrogen peroxide and the second step of not only converting the hydrogen peroxide obtained in the first step to a quinone dye but also treating the remaining low-density lipoprotein to thereby generate hydrogen peroxide and converting the generated hydrogen peroxide to a quinone dye, wherein in the first step no quinone dye is generated and wherein from the amount of quinone dye generated in the second step the cholesterol of low-density lipoprotein and total cholesterol are quantified by one-time measuring.
