PROBLEM TO BE SOLVED: To provide a focusing method for an observation device with which focusing for fluorescent observation is easily performed in a short time. SOLUTION: The focusing method applied to a microscope realizing the observation of fluorescence emitted from a sample 7 includes a 1st step where the sample 7 is irradiated with light through an objective 6 to generate reflected light and the fluorescence, a 2nd step where focusing on the surface of the sample 7 is performed with the reflected light from the sample 7 and a 3rd step where focusing on the fluorescence is performed based on the result of focusing on the surface of the sample 7 in the 2nd step.
