<p><P>PROBLEM TO BE SOLVED: To provide a method for shell-free culture of an embryo of birds with which the embryo in the blastodermic phase of the birds after oviposition can be cultured and developed to result in embryogenesis. <P>SOLUTION: The embryo of a chick in the blastodermic phase shortly after the oviposition is transferred to an oval artificial vessel somewhat larger than the volume of a seed egg and having an opening at right angles relatively to the major axis. An aqueous fresh albumen is filled to the opening which is then hermetically sealed with a plastic film made of a hydrophobic fluororesin as a raw material. The embryo is cultured in an incubator set at 38°C and 60% relative humidity with the opening upward so that the embryo is always brought into contact with film for 72 h. During the culture, the egg is turned at 45-90°angle at a time at an interval of 7.5 min to 71 h. After culture for 72 h, the embryo is developed to stages 17-19 of development stage tables by Hamburger & Hamilton. <P>COPYRIGHT: (C)2006,JPO&NCIPI</p> |
